The factors influencing the development of TN1 arabica coffee somatic embryos cultured in Bioreactor - RITA system

The research was carried out in order to determine the influences of some factors on the development of TN1 arabica coffee somatic embryos cultured in bioreactor - RITA system. TN1 is the F1 generation of hybridizing KH3-1 x Catimor. Four research contents including: 1) Identify the effect of medium components on growth of cotylendon embryos; 2) Examine the time intervals of air supply affecting cotylendon embryos; 3) Determine the influence of nutrition solution volume on the growth of cotylendon embryos; 4) Identify the optimum pH for the development of cotylendon embryos were carried out on the new cultivar TN1 of Arabica coffee. The result indicated that the RITA system with MS medium added saccharose concentration of 10 g/l, air supplying by aeration in one minute for every 6 hours, medium volume of 300 ml for each RITA unit, and pH 5.8 gave the best performance of cotylendon somatic embryos.
