Biological activity evaluation of flavonoids from the EtOAc extract of the leaves of Garcinia mackeaniana

Seven flavonoid compounds isolated from the EtOAc extract of Garcinia mackeaniana leaves include 2”,6”-di-O-acetylvitexin (1), 2”-O-acetylvitexin (2), vitexin (3), amentoflavone (4), apigenin (5), kaempferol (6), and quercetin (7) were evaluated for biological activities. Results showed that both compounds (6) and (7) exhibited strong antioxidant activity with IC50 values of 9.5±0.38 and 7.4±0.30 µg/ml, respectively. Compound (6) had a moderate inhibitory effect on NO production on RAW 264.7 cells (IC50 of 63.86±2.86 µg/ml) with a survival rate of 81.84% at 128 µg/ml. Compound (3) exhibited cytotoxic activity against cancer cell lines MCF7 and LU-1 with IC50 values of 8.0±0.2 and 9.68±0.5 µg/ml, respectively. Compounds (5) and (7) demonstrated strong activity on KB and HepG2 with IC50 values of 8.5±0.35 and 6.79±0.2 µg/ml. Compounds (1-7) were non-toxic on normal cell lines HEK-293 and were inactive against gram-positive test strains.
