Optimisation of direct compression tablet formulation containing spray-dried extract of Eurycoma longifolia Jack

This study aimed to develop a formula for direct compression tablets containing spray-dried extract of Eurycoma longifolia Jack. The flowability of E. longifolia extract and the effect of filler excipients on the flowability of dry extract of E. longifoliaat different ratios was investigated and evaluated. The experimental model consists of 10 formulas designed according to the D-optimal model. The independent variables surveyed included: Starch 1500, A-tab, and croscarmellose sodium (Vivasol GF) ratios. The dependent variables included tablet weight variation, friability, hardness, and disintegration. BC PharSoft OPT software was used to study cause-and-effect relationships and optimise formulas. The optimal formula was experimentally verified and the experimental results were compared with the predicted results using SPSS software. The results showed that all three fillers Starch 1500, A-tab, and Startab could improve the flowability of E. longifolia extract. The optimised E. longifolia tablet formulation included 150 mg of E. longifolia dry extract (30% of tablet weight), 20 mg of Vivasol GF (4% of tablet weight), 212.5 mg of Startab (42.5% of tablet weight), 15 mg of A-tab (3% of tablet weight), 100 mg of Starch 1500 (20% of tablet weight), and 2.5 mg of magnesium stearate (0.5% of tablet weight). The differences between the three experimental results and the predicted results were not statistically significant. This study
has successfully developed the E. longifolia tablet formula and provided important data for the development of the E. longifolia tablet produced by the direct compression method.
