Research on the preparation of microspheres containing bovine serum albumin microspheres from alginate and chitosan

Microspheres are controlled-release drug delivery systems with a micron diameter with a polymeric structure. Alginate and chitosan are two pharmaceutical biodegradable polymers applied for various microsphere preparation techniques. However, there is very limited research on microspheres from alginate and chitosan in Vietnam. Thus, the goal of this research was to prepare and optimise the alginate microspheres containing the model protein bovine serum albumin (BSA) preparation process and then develop a chitosan coating method. The developed method of alginate microspheres containing BSA includes alginate, BSA, isooctane, span 80, tween 80, and calcium chloride. Formulations and process parameters influencing the alginate microsphere properties were optimised using Design Expert v12.0 software. Alginate microspheres containing BSA were coated with 1% chitosan in 1% acetic acid and achieved the controlled release of BSA from microspheres in 24 hours. Chitosan-coated alginate microspheres containing BSA had an average particle diameter of 178 µm, in which the drug loading rate was 12.7%. This study successfully prepared the controlled-release microspheres containing BSA from alginate and chitosan polymers.
