Effects of nutritional components on mycelial biomass and polysaccharides production of Ganoderma australe in submerged culture

Ganoderma is a medicinal mushroom containing bioactive compounds that are beneficial to human health. Polysaccharides extracted from Ganoderma have attracted increasing attention due to their diverse biological activities and potential applications in pharmaceuticals, cosmetics, functional foods, and biotechnology. This study aimed to investigate the effects of different nutrient sources on mycelial biomass production and the accumulation of extracellular polysaccharide (EPS) and intracellular polysaccharide (IPS) of Ganoderma australe (G. australe) in submerged culture conditions. The results showed that, among the carbon sources evaluated, glucose resulted in high mycelial biomass and the highest IPS production with values of 7.65 g/l and 125.07 mg/l, respectively. Among the nitrogen sources evaluated, peptone was identified as the most suitable source, yielding mycelial biomass, EPS, and IPS with values of 7.05 g/l, 74.90 mg/l, and 697.04 mg/l, respectively. For phosphorus sources, KH2PO4 was found to be the optimal source, with mycelial biomass, EPS, and IPS with values of 7.60 g/l, 186.00 mg/l, and 601.14 mg/l, respectively. These findings highlight the importance of nutrient source selection in enhancing mycelial biomass production and polysaccharides accumulation in G. australe in submerged culture.
