The formation of Lam Dong Taxus wallichiana Zucc. callus and cell suspension culture for paclitaxel biosynthesis

Taxus wallichiana Zucc. callus was formed from stem of young tree in B5 medium supplemented with sucrose 20 g/l, 2.4-D 4 mg/l, kinetin 1 mg/l and AgNO3 150 mg/l. The using of 2.4-D 0.1 mg/l, 6-BA 0.5 mg/l and NAA 0.5 mg/l in order to replace the combination of 2.4-D 4 mg/l and kinetin 1 mg/l was suitable for the growth and paclitaxel biosynthesis in callus. Lam Dong Taxus wallichiana Zucc. cell suspension culture made from callus in liquid medium had the same components as in callus culture medium. The growth of cell suspension culture had been affected by initial cell weight and shaking speed. The initial cell weight which was 1-1.5 g of fresh calli in 10 ml liquid medium and shaking speed of 60-110 rpm were suitable for the growth of cell suspension culture.
