Abstract

This study presents the investigation of the conditions affecting Lentinan extraction from Lentinula edodes in Vietnam. RO hot water was used as the extracting solvent, and ethanol 950 was used as a solvent to precipitate recovered product. The scale of the extraction was 20 kg dry material/batch. The authors used the phenol-sulfuric acid method to determine the content of Lentinan (β-glucan) based on the absorption at the 490 nm wavelength of the complex formed by phenol and carbohydrate. The results indicated that the appropriate conditions for extracting Lentinan from dried Lentinula edodes were as follows: dried Lentinula edodes:water (RO) = 1:2.5 (w/v), temperature of solvent (RO water): 1000 C, time of extraction: 80 minutes, extract:ethanol 95= 1/2 (v/v).
